What is Synvisc-One?

Sanofi Genzyme manufactures Synvisc-One®, an injectable gel for the treatment of knee osteoarthritis. It is injected directly into the knee joint to provide pain-relief and lubrication for up to 6 months.

Sanofi Recalls Contaminated Arthritis Injections

In December 2017, Sanofi announced a voluntary recall for over 12,000 syringes of Synvisc-One® arthritis gel injections after an outbreak of serious bacterial infections.

The FDA received dozens of reports of patients who suffered knee infections from Synvisc-One, as well as other serious adverse events.

The recall involves Synvisc-One injections from Lot Number 7RSL021, which were distributed in 36 states (only in the United States) between October 25, 2017 and November 7, 2017.

Synvisc-One Side Effects

  • Infection
  • Pain
  • Trouble walking
  • Unable to put weight on the knee
  • Swelling
  • Fluid build-up in and around the knee
  • Hospitalization
  • Death

Adverse Events Linked to Synvisc-One Injections

A few days before the contaminated Synvisc-One injections were recalled, Sanofi issued an urgent warning to doctors about “an unexpected increase in the number of labeled adverse events.”

For example, in one adverse event, a doctor had to drain fluid from a patient’s knee twice. The patient also suffered continued knee pain.

Sanofi Finds Bacteria in Synvisc-One

The recalled Synvisc-One injections tested positive for “a common airborne microbe” that rarely causes infections in people, according to Sanofi. In a Class II recall, Sanofi identified the microbe in Synvisc-One as Methylbacterium thiocyanatum, which is commonly found in soil, water, or sewage.

Risk of Antibiotic-Resistant Knee Infections

The gram-negative bacteria in Synvisc-One is slow-growing and resists chlorine sanitization processes, so it can potentially grow in syringes for weeks or months before it is detected.

Furthermore, infections are often highly resistant to antibiotics, which significantly increases a patient’s risk of suffering long-term side effects, such as chronic pain.

Treatment for Synvisc-One Infections

The symptoms of a knee infection from Synvisc-One will likely appear within days, but knee problems after an infection can last for months or years.

A patient may need to have a procedure called a “joint aspiration,” in which a doctor uses a needle to drain fluid out of the knee joint. The patient may also need anti-inflammatory corticosteroid injections, antibiotics, or surgery to flush out the contaminated Synvisc-One gel.
