What is Syfovre?
Syfovre (pegcetacoplan injection) is a medication that is injected directly into the eyes of patients who are suffering from geographic atrophy, which is an eye disease that causes patchy lesions.
Syfovre was created by the Massachusetts-based drug-maker Apellis Pharmaceuticals. The company received FDA approval for Syfovre in February 2023, making it the first treatment for geographic atrophy.
FDA Updates Syfovre Warnings for Retinal Vasculitis
In November 2023, the FDA updated the “Warnings and Precautions”, “Postmarketing Experience”, and “Patient Counseling Information” sections of the Syfovre labeling to include retinal vasculitis and/or retinal vascular occlusion.
What is the Problem?
During the first year that Syfovre was on the market, more than 160,000 doses were distributed to physicians. Unfortunately, some patients who received injections of Syfovre developed severe eye inflammation. There were also a handful of reports of occlusive retinal vasculitis, which is a severe eye disease that can cause blindness.
What is Occlusive Retinal Vasculitis?
Occlusive retinal vasculitis is an eye disease that occurs when severe inflammation blocks the flow of blood through vessels that supply the retina. Left untreated, it can cause permanent vision loss or blindness.
Signs & Symptoms of Retinal Vasculitis
Retinal vasculitis may show any of the following symptoms:
- Blurry vision
- Flashes
- Floaters
- Scotoma
- Altered color vision
- Pain
- Visual defects or distorted vision
Letter From Eye Experts Warns of Syfovre Risks
In January 2024, the American Society of Retinal Specialists (ASRS) issued a warning about reports of retinal vasculitis within 1-2 weeks of a patient’s first injection of Syfovre. There were also reports of severe eye inflammation.
According to the ASRS, “fourteen eyes of 13 patients have been confirmed to have retinal vasculitis by review of imaging studies. All cases occurred after the first [Syfovre] injection.”
Apellis Safety Warning for Needle Kits
Apellis has warned healthcare professionals about a risk of “rare safety events” that may be associated with “variations” in the needle kits for Syfovre. Apellis warned doctors to immediately stop using kits with a 19-gauge filter needle, and instead only use kits with an 18-gauge needle.
