Study Links Semaglutide and NAION
The study was published in JAMA Ophthalmology by researchers at Massachusetts Eye and Ear who were investigating cases of an eye disease called NAION in patients on semaglutide.
What is the Risk?
The researchers found 17 cases of NAION in diabetic patients at their institution who were taking semaglutide, compared to 6 cases in diabetic patients who were not taking semaglutide. The study linked semaglutide with a 4.28-fold increased risk of NAION.

What is NAION?
NAION is also called Non-Arteritic Anterior Ischemic Optic Neuropathy, and it is essentially an “eye stroke” that occurs when a lack of blood-flow damages the optic nerve.
What Happens?
The optic nerve is the connection between the brain and the eye. When it is damaged due to NAION, patients may experience sudden, painless vision loss in one eye, often noticed first thing in the morning. This is due to a lack of blood-flow which causes swelling of the optic nerve. The swelling goes down, but the vision loss does not go away.
No Medications Can Cure NAION Blindness
Nerve damage is permanent, which is why vision loss due to NAION is usually incurable. The vision loss may worsen over the next few weeks, but it does not get better. The only treatment involves therapies targeted toward risk-factors for NAION, such as reducing high blood pressure or treating cardiovascular disease and diabetes.

- Sudden painless vision loss in one eye
- Vision loss
- Darkened area of visual field
- “Shadow” over upper or lower half of visual field in one eye
- Loss of contrast
- Blindness
- Doctors may notice swelling of the optic nerve
