FDA Evaluating Rhabdomyolysis Risk
The FDA is evaluating a potential signals of a serious risk and new safety information linking DPP4 diabetes drugs and rhabdomyolysis. This potentially life-threatening kidney side effect was identified by the FDA Adverse Event Reporting System (FAERS) between April and June 2017.
Oseni and Pancreatic Cancer
Oseni (alogliptin + pioglitazone) works by mimicking hormones that cause the pancreas to secrete more insulin, which helps lower blood-sugar levels. Because it relies on the pancreas to work, experts are concerned about pancreatic side effects from Oseni.
The problem is that low-level pancreatitis may go unnoticed for months or years. Over the long-term, chronic inflammation increases the risk of DNA mutations that cause cancer. Oseni belongs to a class of diabetes drugs that have been linked to the following pancreatic side effects:
- Chronic pancreatitis
- Inflammation in the pancreas
- Hemorrhagic pancreatitis
- Necrotizing pancreatitis
- Pancreatic cancer
- Pre-cancerous growths (pancreatic duct metaplasia)
Pancreatic Cancer
Pancreatic cancer is relatively rare, but it is very deadly. The pancreas is located deep inside the abdomen next to many vital organs and lymph nodes. Cancers in the pancreas usually grow aggressively and do not produce symptoms until after it has spread to other organs (metastasized).
FDA Investigates Other Safety Risks
The FDA issued a Drug Safety Communication for Oseni in March 2013, after a study found evidence of pre-cancerous cellular changes. Researchers autopsied pancreases from people who used sitagliptin or exenatide, which are two other drugs in the same class as Oseni.
These medications were also linked to a doubled increased risk of pancreatitis, according to the FDA. In 2010, a study published in Gastroenterology also found higher rates of pancreatitis and pancreatic cancer.
