Kombiglyze and Pancreatic Cancer
AstraZeneca’s Kombiglyze XR is an extended-release diabetes drug that combines metformin and saxagliptin. It is in a class of drugs called “incretin mimetics,” which are being investigated for their possible link to pancreatitis (pancreatic inflammation) and pancreatic cancer.
FDA Safety Warnings
The FDA first warned about the risk of pancreatitis from incretin mimetics back in 2009. Since then, evidence has continued to increase as new drugs in this class have been approved, including Kombiglyze.
In March 2013, the FDA issued a Safety Communication to warn about a study linking two incretin mimetic diabetes drugs with pre-cancerous pancreatic tumors.
The researchers warned about a risk of surgery to remove tumors:
“Since the standard of care of a pancreatic neuroendocrine tumor, because of the risk of conversion to malignancy, even if benign, is surgical resection [i.e. removal], patients exposed to incretin therapy would seem to be at increased risk of requiring pancreatic surgery.”
What is Pancreatic Cancer?
Pancreatic cancer is a rare but often deadly cancer of the pancreas, which is a gland located deep inside the abdomen responsible for producing digestive enzymes and hormones like insulin. Cancer occurs when cells start growing uncontrollably and form a mass called a tumor.
Symptoms of Pancreatic Cancer
Pancreatic cancer rarely produces symptoms in its early stages. Most people are not diagnosed with pancreatic cancer until after it is too late to cure, which is why it is so deadly. Fewer than 5% of people who are diagnosed with pancreatic cancer survive more than five years.
When symptoms of pancreatic cancer do appear, they may include:
- Unexplained fatigue or weakness
- Unintended weight-loss
- Bloating or indigestion
- Sudden worsening of type-2 diabetes
- Abdominal pain
- Nausea or vomiting
- Dark-colored urine, light-colored stools
- And more
